Ambros Therapeutics has closed a $125 million Series A round in December 2025 to build a pipeline of brain-penetrant small molecule therapies for neuropsychiatric and neurodegenerative diseases. The round is one of the largest Series A financings in CNS drug development history.
The Challenge
Developing drugs for brain disorders has historically been plagued by two fundamental challenges: the blood-brain barrier blocks most drug candidates from reaching their target, and the complexity of brain circuitry makes it difficult to predict which molecular targets will produce therapeutic effects without unacceptable side effects.
Ambros's Approach
The company's platform combines computational chemistry with advanced pharmacology to design small molecules that efficiently cross the blood-brain barrier and selectively engage specific neuronal targets. Their approach emphasizes 'circuit selectivity' — targeting molecular mechanisms within specific brain circuits rather than broadly modulating neurotransmitter systems.
Pipeline
While specific programs remain undisclosed, Ambros has indicated its pipeline spans treatment-resistant depression, cognitive impairment in schizophrenia, and early-stage neurodegeneration — conditions where current therapies are inadequate and the unmet medical need is enormous.
