Barrier tissues-including the skin, respiratory tract, and gastrointestinal tract-are dynamic interfaces where neural and immune systems converge to coordinate barrier defense, inflammation, and repair. Recent studies show that neurons and immune cells form bidirectional networks that sense environmental cues, pathogen-derived signals, and tissue stress to fine-tune immunity and maintain homeostasis. Sensory, sympathetic, parasympathetic, and enteric neuronal pathways shape epithelial function, vascular dynamics, and immune activation across barrier tissues. Through neurotransmitters and neuropeptides, neuroimmune circuits regulate pathogen clearance, preserve tissue integrity during infection, modulate allergic and autoimmune responses in sterile inflammation, and support tissue regeneration by instructing immune cell reprogramming and epithelial renewal.
